Stable isotope and multi-element profiling of Cassiae Semen tea combined with chemometrics for geographical discrimination

摘要: Cassiae Semen (Sicklepod seeds), a popular Chinese traditional medicinal tea that is roasted, crushed, and infused in boiled water to make a commonly used traditional tea drink. The quality and efficacity of Cassiae Semen are closely related to its geographical origin. Therefore, stable isotope (delta C-13, delta N-15, delta H-2, and delta O-18) and multi-element (32 elements) chemometrics were used to develop a geographical discrimination method for Cassiae Semen. In this study, significant isotopic and elemental differences (p < 0.05) were found for Cassiae Semen teas from different growing regions in China and Vietnam. Regional samples were separated using unsupervised PCA, and OPLS-DA identified specific elemental and isotopic variables (K, Ca, Mg, Al, Fe, Ba, and delta H-2) as key origin discrimination markers. OPLS-DA had a high discrimination accuracy rate (100 % and 99.6 %) for high-quality Cassiae Semen tea cultivated in Xinchang County, Zhejiang Province (ZJXC), China, and Hanoi Province, Vietnam (VIEHN), respectively. Multi-element and stable isotope analyses combined with chemometrics provide a promising method to confirm the geographical origin of Cassiae Semen.
