Macrophage-inspired magnetic silk fibroin porous microcarriers with exosome enrichment capability for bone repair

摘要: Bone defect remains a major challenge for clinicians. Exosome (Exo) therapy is a promising approach for bone repair. The current trend of Exo therapy is to develop multifunctional Exo delivery microcarriers to promote bone repair. Here, inspired by macrophage phagocytosis, magnetic silk fibroin porous microcarrier (MS) with Exo enrichment ability is developed via microfluidic technology for bone repair. Benefiting from the good droplet manipulation ability of microfluidic technology, the generated MS have good monodispersity and spherical morphology. With the assist of liquid nitrogen pre-freezing and ultraviolet (UV) crosslinking, the produced MS showed porous structure. Additionally, the addition of Fe3O4@MgSiO3 nanoparticle and the porous structure endowed the MS with good magnetic properties and satisfactory Exo capture capability after antibody modification. Moreover, the released Exo and microelement of Fe3O4@MgSiO3 from MS can effectively improve bone repair via angiogenesis and osteogenesis. The cell and animal experiments demonstrated that this kind of MS microcarriers have good biocompatibility, Exo enrichment ability and bone repair effect. These characteristics indicate that the designed MS is an effective carrier for Exo enrichment and delivery, and is expected to be a viable option for clinically relevant diseases therapy.
