Biotransformation of inorganic selenium into selenium nanoparticles and organic selenium by Lactiplantibacillus plantarum CXG4

摘要: Selenium enrichment by microorganisms is a promising strategy in the emerging selenium industry, wherein inorganic selenium (Se(IV)) is converted into organic selenium and nanoselenium. This study demonstrated that Lactiplantibacillus plantarum CXG-4 could effectively transform Se(IV) into organic selenium and selenium nanoparticles (SeNPs). Under optimal culture conditions, the selenium enrichment rate and selenium content of L. plantarum CXG-4 reached 64.11% +/- 0.90% and 1874 +/- 14 mu g/g, respectively. SeNPs were the major selenite reduction products (95.02%) from L. plantarum CXG-4, and the organic selenium that primarily existed in the form of proteins constituted 4.98% bound in the total biotransformed selenium. X-ray photoelectron spectroscopy revealed the successful transformation of selenium and the participation of sulfur in the reduction of selenium. XRD analysis indicated that the SeNPs exhibited an amorphous morphology. Fourier-transform infrared spectroscopy showed that selenium combined with protein or polysaccharides to form organic selenium. Transmission electron microscopy and scanning electron microscopy showed that the diameters of SeNPs ranged from 35 to 240 nm. Furthermore, whole-genome analysis showed that L. plantarum CXG-4 had various genes related to Se(IV) reduction, such as thioredoxin family protein genes and some short-chain dehydrogenase reductase family oxidoreductase genes. Remarkably, transcriptome and real-time quantitative PCR analyses further validated that L. plantarum CXG-4 could reduce Se(IV) by upregulating the expression of the related genes trxA, adhE, mtlA, and msrA. These results provide novel insights into the transformation of Se(IV) by lactic acid bacteria, which is important for the development of novel selenium nutritional supplements.
