Heavy metals and bacterial community determine resistance genes distribution in agricultural soils surrounding long-term mining area

摘要: The spread of antibiotic resistance genes (ARGs) and metal resistance genes (MRGs) has become a huge threat to public health. However, the interactions between resistance genes and heavy metals and bacterial community in agricultural soils around mining areas remain unclear. In this paper, agricultural soils around a long-term mining area in eastern China were analyzed to clarify the MRG and ARG profiles and bacterial community composition using metagenomics technology, and chemical properties and heavy metals were considered as environmental factors. The results showed that genes resistant to Cu, As, and multi-metals were dominant MRGs, and arsM was the most abundant subtype. The main ARGs conferred resistance to peptide and rifamycin antibiotics, with the most abundant subtype being rpoB2. PLS-PM analysis revealed that the ARG profiles were positively and directly controlled by the bacterial community, whereas the MRG profiles were jointly shaped by heavy metals, chemical properties, and bacterial community, in which heavy metals contributed the greatest total effect. Co-occurrence network analysis demonstrated that the bacterial genera potentially carrying more ARG subtypes mostly belonged to Proteobacteria, including Paraburkholderia, Azospirillum, Polaromonas, and Cupriavidus. The potential hosts for MRG subtypes were mainly sulfur- or iron-related bacterial genera, including Sulfuricaulis, Sulfurifustis, Thiobacillus, Sideroxydans, and Anaeromyxobacter. This study emphasized different roles and regulating pathway of heavy metals and bacterial community in resistance genes distribution in agricultural soils under long-term heavy metal stress, which is conducive to developing targeted strategies for the risk management in contaminated areas.
