Challenges in the characterization of pectin conformation: Integrated analysis using atomic force microscopy and small-angle X-ray scattering methods

摘要: Pectin is a complex, naturally heterogeneous polysaccharide, exhibiting specific physicochemical properties and a wide range of physiological activities. Although a growing number of studies have demonstrated that the conformation of pectin plays a crucial role in its properties and functions, conformational studies remain methodologically challenging. This review examines the principles, methodology, and prior applications of atomic force microscopy (AFM) and small-angle X-ray scattering (SAXS) in studies of pectin conformation. The advantages and disadvantages of two methods, both alone and in combination, and trends in this field are also discussed. AFM allows imaging of the single molecular chains of pectin and thus provides conformational and size information; however, it requires a dry sample environment. The use of mathematical models to interpret scattering curves obtained by SAXS yields in situ information on the conformation of pectin aggregates. By integrating SAXS and AFM results, structural information can be obtained from mathematical models and visualizations, including the structural scale of pectin in various environments. Our review will contribute to improved conformational characterizations of pectin and thus to an elucidation of its structure-function relationships.
