Screening of raw materials for antifreeze protein extraction and comprehensive analysis of their regulatory networks

摘要: Antifreeze proteins (AFPs) are a class of specialized proteins capable of inhibiting ice crystal formation and growth in extremely low-temperature environments. AFPs have tremendous potential applications in various fields such as medicine, industry, and agriculture. As natural metabolic products, AFPs are eco-friendly and have potential as preservatives, medical cryopreservation alternatives, and road salt substitutes. Plant AFPs, possessing both antibacterial and antifreeze properties, offer additional advantages. Due to the antifreeze proteins in plants having both antibacterial and antifreeze properties, they offer additional advantages. To identify suitable sources for extraction, we screened three overwintering crops and found that Brassica napus showed a higher number and abundance of antifreeze protein gene expression compared to the other two species. To further enhance the expression abundance, we analyzed the expression patterns of antifreeze genes in Brassica napus and discovered a potential relationship between regulatory patterns and expression levels. By integrating Brassica napus Hi-C gene interaction database with the transcription factor database, we predicted transcription factors that regulate the highly expressed antifreeze genes and the significantly differentially expressed antifreeze genes under cold stress.Thus, a potential regulatory network of antifreeze genes was established. We also quantified the number of transcription factor binding sites and found that the top three were GRAS, MIKC_MADS, and NAC. Notably, the binding site of the transcription factor BnaC04g26350D, which regulates the BnaC04g24330D gene, is MIKC_MADS. These findings provide a theoretical foundation for increasing the expression abundance of AFP genes.
