High Frequency Adventitious Shoot Induction and Plant Regeneration from Hypocotyl of Ramie [Boehmeria nivea (L.) Gaud]

摘要: An efficient regeneration system for adventitious shoot induction was developed using hypocotyl explants of ramie [Boehmeria nivea (L.) Gaud], either by shoot organogenesis or direct regeneration of shoots, without any intermediate callus phase. The regeneration efficiency depended on the concentrations of plant growth regulators. Shoot induction directly from hypocotyl explants with high frequency (34%) was obtained on Murashige and Skoog (MS) medium supplemented with 1.11 mu M benzyladenine (BA) and 2.27 mu M Thidiazuron (TDZ). Furthermore, efficient indirect regeneration occurred when hypocotyls were first plated on MS medium supplemented with 2.22 mu M BA and 1.43 mu M indoleacetic acid (IAA), and then transferred to shoot-inducing medium containing 0.045 mu M TDZ, 0.69 mu M IAA and 0.35 mu M gibberellic acid (GA(3)); the frequency of shoot regeneration was 82%. The regenerated shoots were rooted on a root-inducing medium containing 2.46 mu M naphthaleneacetic acid (NAA) with 100% rooting frequency. The results demonstrated efficient adventitious shoot induction and plant regeneration of ramie. This protocol provides a new biotechnological strategy to transfer economically important genes to this important crop species.
