Genome-Wide Identification of Expansin Gene Family and Its Association with Sepal Expansion During Flower Opening in Hydrangea macrophylla

摘要: Hydrangea macrophylla is a widely cultivated ornamental plant and renowned for its colorful and bountiful floral inflorescences. Sepals, rather than petals, are the primary ornamental structures of Hydrangea. Expansin genes play critical roles in flower opening, however, their regulation in the growth and development of sepals in Hydrangea remains unclear. In this study, 26 expansin genes were identified based on genome and transcriptome data in Hydrangea, and they were classified into four subfamilies (EXPA, EXPB, EXLA and EXLB). Across different species, gene structure and conserved motifs of EXPs exhibited evolutionary conservation. The promoters of Hydrangea expansin genes primarily contained four categories of cis-acting elements: abiotic stress response, environment response, phytohormone response and transcription factor binding sites. Furthermore, HmEXPA4, HmEXPA15, and HmEXPA17 were found to be associated with growth and development of sepals in decorative flowers, while five transcription factors (HmCIB1, HmPIF8, HmIAA14, HmIAA16, and HmHB1) were identified as potential upstream regulators of these HmEXPs. This research provides a foundation for the further studies on the mechanism underlying sepal development in Hydrangeas.
