Establishment of recombinant doubled haploid lines of Japanese flounder and evaluation of their reproductive capacity

摘要: Doubled haploids are assumed to be free of lethal recessive and major deleterious alleles, and are thus appropriate for use in selective breeding programs for improving fish stocks. However, in the case of the Japanese flounder (Paralichthys olivaceus), an important aquacultural fish, the application of doubled haploids in selective breeding has been severely hampered by the infertility of doubled haploid lines. In this study, we succeeded in enhancing the reproductive traits of recombinant doubled haploids via gynogenesis combined with a targeted breeding technique, thereby obtaining 80% fertile recombinant doubled haploids. The specifics of this approach were as follows: the first generation was established from wild-caught Japanese flounders with superior reproductive traits via meiotic gynogenesis in 2007, and similarly, in 2011, the second generation was established from the first generation with superior reproductive traits via mitotic gynogenesis. In 2015, the third generation was established from the second generation with different genetic backgrounds via hybridization, and in 2019, recombinant doubled haploid lines were established from the third generation with superior reproductive traits via mitotic gynogenesis. Compared with the doubled haploids, 80% of the recombinant doubled haploids were fertile, and the reproductive traits of these recombinants, including fertilization rate, hatching rate, and amounts of artificially squeezed eggs, had all undergone significant increases. Moreover, all recombinant doubled haploids were found to be homozygous, as determined by genotyping using 24 microsatellite markers. In this study, we accordingly developed an effective method for improving the reproductive traits of doubled haploids, using which, we succeeded in producing a large number of fertile recombinant doubled haploids, thereby providing valuable resources for the selective breeding of improved varieties of Japanese flounder.
