Identification of Cucurbit Aphid-Borne Yellows Virus in Cucurbita moschata by Deep Sequencing and Characterization of Virus-Derived Small Interfering RNAs

摘要: Deep sequencing of small RNA populations in cucurbits plants in central China resulted in the identification of the cucurbit aphid-borne yellows virus (CABYV), a member of the genus Polerovirus in the family Luteoviridae. RT-PCR was performed to validate the presence of this virus and the virus-derived small interfering RNAs (vsiRNA) of CABYV in the cucurbits plant were characterized. CABYV-derived siRNAs with length of 21-22 nt were predominated, indicating a possible conserved mechanism of DCL2 and DCL4 participation in the generation of vsiRNAs. A slight preference of C/U towards the 5'-terminal base of CABYV-vsiRNA was observed, suggesting that CABYV-vsiRNA might be loaded by distinct AGO complexes to disturb the gene expression of host plants. The CABYV-vsiRNAs were not evenly distributed along the viral genome, and MP/CP/RTD-coding regions of genome segments were detected in the 5'-terminal regions. Our results might contribute to clarify the host antiviral response induced by CABYV, and give hints on designing antiviral strategies using RNAi against CABYV.
