Vitamin E stimulates the expression of gonadotropin hormones in primary pituitary cells of turbot (Scophthalmus maximus)

摘要: The relationship between vitamin E and gonadotropin hormones (FSH: follicle-stimulating hormone, LH: luteinizing hormone) in turbot was conducted by providing vitamin E through primary pituitary cells in vitro. Pituitaries were sampled aseptically and digested with 0.25% trypsin-EDTA for about 15 min at room temperature, and then were placed into the L-15 Medium, containing 5% fetal bovine serum, 100 U ml(-1) penicillin and 100 mu g ml(-1) streptomycin. After adding vitamin E (DL-alpha-tocopherol acetate from Sigma: 0, 9, 18, 36, 72 and 144 mu mol L-1) into the cells for one day, the cells were separated and collected for the mRNA analysis. Results showed that the mRNA expression of FSH-beta (fsh-b) were significantly increased when they were treated with vitamin E, and the significant increasing trends were dose-dependent with vitamin E concentrations; while the LH-beta (1h-b) mRNA expression were significantly increased with vitamin E from 0, 9 to 18 mu mol L-1 and not significantly increased any more. These data demonstrated that Vitamin E stimulates the expression of gonadotropin hormones in the pituitary of turbot in vitro. It also provides an alternative way to explore the mechanism of nutrient acting on the fish reproduction with this convenient cellular pituitary model.
