Transposon insertion sequencing analysis reveals conditional essential genes for infection in Pseudomonas plecoglossicida

摘要: Pseudomonas plecoglossicida is a critical pathogen that affects cultured large yellow croakers (Larimichthys crocea). While several virulence genes in this bacterium have already been identified, a comprehensive understanding of its pathogenicity is still lacking. To address this gap, we constructed the transposon insertion mutant library for P. plecoglossicida and conducted an in vivo screening of this library for the first time. In this study, we revealed that 251 genes are essential, while 145 additional genes are conditionally essential during the infection process. Furthermore, we proposed that the absence of the mlaE (Delta mlaE) resulted in an attenuated phenotype, emphasizing the ability of Delta mlaE to be a live attenuated vaccine due to its reduced ability to survive under host's antimicrobial defenses. This study employed a powerful tool for functional genomic analysis in P. plecoglossicida, and provided a more holistic view of its pathogenicity. By gaining more insights into the virulence mechanisms of P. plecoglossicida, we can potentially develop more effective therapeutic strategies against its infections.
