Application of reverse transcription loop-mediated isothermal amplification for rapid detection of East Asian passiflora virus (EAPV) in malformed passion fruit

摘要: Passion fruit cultivation in East Asia, most notably in China, faces threats from the East Asian passiflora virus (EAPV), which induces malformation in fruits. The objective of this study was to optimize the reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay for rapid EAPV detection in passion fruit. Fruit samples were collected from Guizhou province, a major passion fruit-producing area in China. Five sets of primers were designed using the coat protein gene (CP) of EAPV as the target sequence and were screened to check amplification efficiency using a real-time fluorometer under various reaction conditions. The optimized RT-LAMP assay achieved higher sensitivity and specificity than reverse transcription-polymerase chain reaction in 90 malformed fruit samples. The optimized RT-LAMP assay is a simple, highly sensitive, and reliable method for rapid visual detection of EAPV in passion fruit. To the best of our knowledge, this study is the first to apply and optimize RT-LAMP for EAPV detection and is expected to support general viral management alongside EAPV-specific control measures through rapid and cost-effective detection for maintaining passion fruit crop health.
