Cytological identification of a genus-wide chromosomal fusion-fission cycles in genus Morus

摘要: As an economically important woody perennial tree, mulberry (Morus spp.) is widely distributed in the world. A dynamic chromosomal fusion-fission cycle has been identified in M. notabilis. However, the prevalence and mechanism of this chromosomal fusion-fission cycle in the genus Morus remain unclear. In the present study, fluorescence in situ hybridization (FISH) was performed in 26 mulberry accessions belonging to 14 species of the genus Morus using 25S rDNA as the probe, which is specifically located at the chromosomal fusion-fission region of M. notabilis. The dynamic signal patterns of the 25S rDNA probe indicated that the chromosomal fusion-fission cycle is common in the genus Morus. Further, because of the similarity in size between chromosomes 4 and 5 in M. notabilis, the largest diakinesis chromosome 1, which is involved in the chromosomal fusion-fission cycle in M. atropurpurea 'Lunjiao109', was selected for chromosomal microdissection. A chromosome-specific DNA library was constructed, and 21 clones containing abundant repeat sequences were obtained. Four FISH signal patterns were obtained using these clones as probes. Additionally, two sequences of the selected clones might be involved in the chromosomal fusion-fission cycle. The present study identifies the chromosomal fusion-fission cycle as a widespread phenomenon throughout the genus Morus. This cycle can provide an excellent vehicle for analyzing the mechanism of chromosomal evolution in mulberry.
