Identification and function analysis of drought-responsive miRNAs in sorghum (Sorghum bicolor)

摘要: Drought is a common abiotic stress which can affect crop quality and limit crop production worldwide. Recently, microRNAs (miRNAs) were reported to play important roles in plant abiotic stress response. Sorghum (Sorghum bicolor) ranks the fifth among grain crops in terms of production and is applied in various fields, such as chemical, brewing, food, bioenergy, and feed industries. While the protein-coding genes related to drought resistance in sorghum have been identified, the drought-responsive miRNAs in sorghum need to be explored. In the current work, three small-RNA libraries were prepared based on plants under three distinct treatments-normal water supply (CK), mild drought stress (DR1), and severe drought stress (DR2). A total of 108 known miRNAs and 14 mature novel miRNAs were identified, among which 17 known miRNAs and 2 novel miRNAs exhibited significant differential expression between the plants under the CK, DR1, and DR2 treatments. Moreover, we predicted 227 target genes of these drought-responsive miRNAs. The Gene Ontology (GO) analysis conducted for the target genes of the drought-responsive miRNAs revealed "nucleobase-containing compound-related metabolic process", "cell component", and "electron transfer activity" as the GO terms with the highest enrichment levels. The above results contributed to research on drought resistance and provided further insight into the drought adaptation and tolerance mechanisms of sorghum.
