Novel antioxidant peptides from hydrolyzed proteins of Volvariella volvacea: Purification, characterization and protective effects on H2O2-induced human skin fibroblasts

摘要: In this study, five novel antioxidant peptides were isolated, purified, identified and characterized from Volvariella volvacea proteins by enzymatic digestion, LC-MS/MS and molecular docking. The results showed that fractions with molecular weights less than 1 kDa from V. volvacea peptides (VP) obtained using neutral proteolysis possessed significant antioxidant capacities. These peptides were purified through ion exchange chromatography and gel chromatography using a Q Sepharose FF column and a Superdex 30 Increase column, respectively. Subsequently, five novel antioxidant peptides (DWPGFK, SFDWTGFK, FDWPGFKT, DWPTFKAF and SGPSFDWPGFK) were identified by LC-MS/MS and bioinformatic analyses, of which, DWPGFK and DWPTFKAF exhibited strong in vitro antioxidant activities. Moreover, these two peptides, especially DWPGFK, protected human skin fibroblast (HSF) from oxidative damage, enhanced antioxidant enzyme activity, and inhibited matrix metalloproteinase activity, thereby preventing excessive collagen degradation and mitigating skin aging. Molecular docking studies revealed that DWPGFK probably exerts its antioxidant effects by activating the Keap1- Nrf2-ARE pathway. This work provided a new breakthrough point and research direction for in-depth exploration of the bioactive components of V. volvacea and the development of their high value-added applications.
