Aronia melanocarpa Elliot anthocyanins inhibit colon cancer by regulating glutamine metabolism

摘要: An azoxymethane (AOM)/dextran sulfate sodium (DSS) mouse model and the Caco-2 cell line were used to evaluate the therapeutic effects and mechanisms of Aornia melanocarpa Elliot anthocyanins (AMA) in colitis associated colorectal cancer (CRC). In this study, AMA could significantly inhibit the proliferation of Caco-2 cells; AMA alleviated colorectal damages caused by AOM/DSS, ameliorated CRC model's body weight fluctuation, reduced the colorectal inflammation histological assessment, decreased CRC related inflammatory cytokines (COX-2, MUC2, MPO, IL-6, IL-17, TNF-alpha and IFN-gamma) and the expression of Glutaminase (GLS) and SLC1A5, downregulated the mTORC1 signaling pathway (p-p70s6k, p70s6k, ULK1, p-mTOR, 4EBP1, p-4EBP1), suggesting that the protective mechanism of AMA for CRC may involve in decreased GLS and SLC1A5 and downregulating the mTORC1 signaling pathway. In addition, the current study provided theoretical support of the mechanism for further colorectal cancer research and provided a new direction for the prevention and individualized treatment of CRC.
