Transcriptomics revealed the regulatory mechanism of phenylalanine butyramide on the growth and biofilm formation of Lactobacillus paracasei

摘要: Phenylalanine butyramide (FBA), a butyric acid derivative, is advantageous in the improvement of intestinal flora. Nevertheless, it remains unclear whether FBA is directly beneficial for lactic acid bacteria. In this study, we investigated the effects of FBA and its synthetic substrates, butyric acid and phenylalaninamide, on Lactobacillus paracasei. Results showed that, contrasted with butyric acid and phenylalaninamide, FBA not only significantly promotes the growth and biofilm formation of L. paracasei but also inhibits the proliferation of pathogenic bacteria. In scanning electron microscopy and in vitro gastrointestinal tolerance assays, FBA elevates the extent of adhesion between biofilm cells and their tolerance to gastrointestinal fluids. Additionally, the comparative transcriptomic analysis revealed that FBA regulates bacterial growth and biofilm formation by upregulating genes associated with histidine biosynthesis, cell division, stress response, and polysaccharide synthesis. This study further suggests that FBA holds promise as an alternative to butyric acid due to its comparable or even better attributes.
