Comparative phenotypical and transcriptional analysis of two contrasting rice genotypes under salt stress

摘要: High salinity poses a significant challenge to the quality and productivity of rice (Oryza sativa L.). While some rice varieties are recognized as salt-tolerant, few with a strong "Simiao rice" background, particularly suitable for production in South China, have been bred in recent years. This study extensively evaluated salt tolerance in two Oryza indica varieties: Nan Gui Zhan (NGZ) and Gui Jin Si Miao (GJSM). We utilized agronomic analysis, experimental treatment, physiological evaluation, and gene variation analysis to comprehensively explore the internal changes between the two rice varieties. Results indicate that GJSM is salt-sensitive, whereas NGZ exhibits salt tolerance. Productivity, grain quality, osmotic pressure content index, and antioxidant enzymes were compared between the two varieties. Furthermore, the novel allele qSKC1(NGZ) was identified, possibly contributing to high salt tolerance with its enhanced expression under salt stress conditions. RNA-seq analysis revealed up-regulation of Na+/K+ transporters, salt-responsive transcription factors, and other salt response-related proteins in NGZ under salt stress, suggesting a potential contribution to more effective Na+/K+ homeostasis in its phenotype. In conclusion, this study offers a thorough examination of the phenotypical and physiological characteristics of two indica varieties. It also delves into an in-depth analysis of the salt tolerance mechanism in rice, providing valuable insights into the molecular breeding of salt-tolerant rice varieties. [GRAPHICS] .
