Metabolome and transcriptome analyses reveal that pollination with 'Guanxi' honey pomelo pollen alleviates the postharvest fruit granulation of 'crystal' honey pomelo

摘要: 'Crystal' honey pomelo is a local special pomelo variety on the border of western Yunnan with an excellent flavor, but granulation is prone to occur in its juice sacs after harvest. This study investigated the effect of crosspollination with 'Guanxi' honey pomelo pollen on the fruit granulation of 'crystal' honey pomelo during storage, aiming to provide a theoretical basis for improving the postharvest quality of 'crystal' honey pomelo fruit. First, 'crystal' honey pomelo was pollinated with pollens from 'Guanxi' honey pomelo. Then, liquid chromatography-mass spectrometry (LC-MS) and RNA seq were used to analyze the differential metabolites and differentially expressed genes (DEGs) in the juice sacs of 'crystal' honey pomelo fruit after cross-pollination treatment (T) with selfpollination as the control (CK) at 0, 50, and 80 d of postharvest storage. The results revealed that obvious granulation occurred in the juice sacs of CK fruit after 80 d of storage, while granulation was significantly delayed in T fruit. Metabolome analysis revealed 42 classes of significantly differential metabolites involving 235 individual metabolites during the three storage periods. Most primary metabolites were significantly upregulated while the majority of secondary metabolites were dramatically down-regulated in the juice sacs of T fruit. Moreover, T fruit showed significant increases in flavonoids such as kaempferol 3-O-robinobioside and lonicerin in the flavonoid metabolism pathway, while significant decreases in phenols in the lignin accumulation pathway, resulting in inhibited lignin accumulation. Transcriptome analysis revealed significant inhibition of phenylpropanoid biosynthesis in the juice sacs of T fruit, and identified 54 DEGs related to lignin accumulation, among which 49 genes (90.7 % of the total DEGs, particularly PAL, C4H, CCR, F5H, and POD) were significantly down-regulated. These results indicate that cross-pollination with 'Guanxi' honey pomelo pollen inhibits lignin accumulation in the juice sacs of 'crystal' honey pomelo, thereby alleviating postharvest juice sac granulation of the fruit.
