Expression Analysis of Low Temperature Responsive Genes in Eupatorium Adenophorum Spreng Using cDNA-AFLP

摘要: Eupatorium adenophorum Spreng, with high ecological adaptation, has been gradually threatening agriculture and forest in various regions. In the present study, to understand the underlying molecular mechanism in response to low temperature, we investigated the expression of genes in Eupatorium AS under low temperature using cDNA-amplified fragment length polymorphism. Across 240 primer recombinants, 125 transcript-derived fragments (TDFs) were screened and sequenced, including the putative genes encoding photosynthesis-related proteins, enzymes, as well as development-related proteins. Sequence alignments showed that several candidate TDFs shared high levels of identity with corresponding genes from Nicotiana tabacum, Oryza sativa, Arabidopsis thaliana, and other species. Furthermore, the inducible expression of these genes under cold type II chlorophyll a/b binding protein and cytosolic phosphoglycerate kinase was confirmed by reverse transcriptase-polymerase chain reaction. These findings will bebeneficial for understanding the underlying physiological, biochemical, and molecular events involving in Eupatorium AS adapting to different environments.
