Genetic Variability of the Mitochondrial DNA D-Loop in Laboratory Closed Colonies of the Golden Hamster (Mesocricetus auratus)

摘要: Closed colonies of the golden hamster (Mesocricetus auratus), a widely used animal model in biomedical research, are expected to have low variability. However, because of their limited genetic background, it remains to be clarified whether or not four golden hamster colonies in China come from the same origin. We analyzed the genetic diversity of the mitochondria! DNA (mtDNA) D-loop sequence of the golden hamsters using single-nucleotide polymorphism (SNP). Based on our results, domain I from the 1 to 420 positions and domain Ill from the 651 to 867 positions were the higher variant regions and domain III from the 421 to 650 positions was the comparatively conserved central segment of the mtDNA D-loop in four golden hamster colonies from China. Ninety-five golden hamsters from four closed colonies were found to be distributed in 58 haplotypes. H1 haplotype was shared by all four closed colonies of golden hamsters and H53 was shared by the three closed colonies (Lanzhou, Beijing and Shanghai). The four closed colonies of golden hamsters shared a common maternal lineage while each colony had its own specific characters. These results provide information on the distinct genetic background of golden hamsters.
