A Novel DNA Extraction Method for the Detection of Jujube Witches' Broom Phytoplasmas

摘要: The cultivar 'Jinsi 4' of Chinese jujube was used to establish a novel DNA extraction method for the detection of jujube witches' broom phytoplasmas. The result showed that the amplified fragment from the crude DNA extract of jujube JWB-infected leaves was the same as that from the total DNA extract using the conventional improved SDS-CTAB method, which was classified as a member of 16SrV-B subgroup. Then the cultivars 'Yuanling 1', 'Lubeidongzao' and 'Dabailing'of Chinese jujube were used to verifythe applicability of the established novel DNA extraction method, the results showed that these 3 isolates were also classified as members of 16SrV-B subgroup. And the online virtual 16S rDNA-RFLP analysis showed that these 4 isolates (accession No.:HM989945, HM989946, HM989947 and HM989948) shared high identity with JWB-G1 (AB052876), their similarity coefficient were 0.98, 0.97, 0.98 and 0.98 respectively, which indicated that the novel DNA extraction method established in this study was practically used to detect jujube witches' broom phytoplasmas.
