Promising MS2 mediated virus-like particle vaccine against foot-and-mouth disease

摘要: Foot-and-mouth disease (FMD) has caused severe economic losses to millions of farmers worldwide. In this work, the coding genes of 141-160 epitope peptide (EPi4i_i6o) of VP1 were inserted into the coat protein (CP) genes of MS2 in prokaryotic expression vector, and the recombinant protein self-assembled into virus-like particles (VLP). Results showed that the CP-EP141_160 VLP had a strong immunoreaction with the FMD virus (FMDV) antigen in vitro, and also had an effective immune response in mice. Further virus challenge tests were carried out on guinea pigs and swine, high-titer neutralizing antibodies were produced and the CP-EP14i_16o VLP vaccine could protect most of the animals against FMDV. Foot-and-mouth disease (FMD) has brought severe economic losses to stockbreeding and it continues to be a serious threat (Sobrino et al., 2001; Pengyan et al., 2008). Vaccination and slaughter of the infected or exposed herds are the main measures taken to control outbreaks of FMD. Conventional inactivated vaccine has played a leading role in controlling epidemics (Chen et al., 2006, 2004; Wang et al., 2002). However, it also presents the risk of virus escaping from production facilities (Strohmaier et al., 1982; Guo et al., 2005; King et al., 1981). Besides, it is hard to distinguish carrier animals from uninfected vaccinated animals (Wang et al., 2002). Moreover, its thermal instability, short protection period and other hazardous factors make the traditional vaccine far from optimal (Wang et al., 2002; Shao et al., 2011). Thus, it is urgent to develop alternative effective and safe vaccines. FMD virus (FMDV), the etiological agent of FMD, has seven distinguishable serological types and more than 70 subtypes (Domingo et al., 2002; Bachrach, 1968; Knowles and Samuel, 2003).
