Bioinformatic identification and expression analysis of the Malus domestica DREB2 transcription factors in different tissues and abiotic stress

摘要: DREB transcription factors regulate the expression of many abiotic-stress-responsive genes and play critical roles in improving abiotic stress tolerance in plants. In this study, the MdDREB2 transcription factors in apple (Malus domestica) were identified through bioinformatics approach, and MdFREB2 transcript levels in different tissues and abiotic stress conditions were also further studied using RT-PCR methods. Fifteen MdDREB2 transcription factors were identified in apple genome, and all contain NLS motif and AP2 domain. The phylogenetic analysis showed that MdDREB2 transcription factors were divided into 3 subtypes: subtype 1, 2, and 3. Subtype 1 included MdDREB2A, B, D, O, L, and subtype 2 had MdDREB2 N and MdDREB2M, while MdDREB2F, G, K, J, H, I, E belonged to subtype 3. Chromosome mapping analysis showed that the largest number of MdDREB2 transcription factors were found on chromosome 12 (7 genes), followed by chromosomes 1, 4 and 8 (2 genes each), and only one gene was located on chromosomes 7 and 17, respectively. Quantitative RT-PCR results demonstrated that MdDREB2 transcription factors had similar expression patterns and had higher expression levels in roots and leaves than other tissues. Besides, their expression level was increased more or less under drought and salinity stress. In summary, MdDREB2 transcription factors shared highly conserved protein structure, and were responsive to abiotic stresses, which indicated the important role against various environmental conditions.
