Control of post-harvest soft rot (Rhizopus stolonifer) on sweet cherry using volatile organic compounds produced by Bacillus velezensis Q-84

摘要: The soft rot of sweet cherries caused by Rhizopus stolonifer (R. stolonifer) results in significant economic losses during storage and transportation. This study demonstrated that volatile organaic compounds (VOCs) derived from the fermentation broth of Bacillus velezensis (B. velezensis) Q-84 significantly inhibited the growth of R. stolonifer by 54.43%. Scanning electron microscope (SEM) micrographs of VOCs-treated fungi revealed antifungal activity, evidenced by the breakage and deformation of the mycelium. Through headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC/MS) analysis, ten VOCs were identified in the fermentation broth of B. velezensis. Notably, 5-methyl-2-heptanone, 2,5-dimethylpyrazine, 2-decanone, and benzaldehyde exhibited significant antifungal activity in vitro. Furthermore, in vivo assays demonstrated the efficacy of both individual and combined VOCs in managing soft rot diseases in cherries. These treatments also resulted in increased levels of total polyphenolic content (TPC) and total flavonoid content (TFC), as well as a significant enhancement in the activities of defense-related enzymes such as peroxidase (POD), polyphenol oxidase (PPO), phenylalanine ammonia-lyase (PAL), and chitinase (CHI). Our results further our understanding of B. velezensis antifungal metabolites on the management of R. stolonifer and may help in finding less toxic compounds to control soft rot on sweet cherries.
