Determination of Phenylethanolamine A in Animal Feeds Using Ultra High Performance Liquid Chromatography-Photodiode Array Detector

摘要: Ultra high performance liquid chromatography with a photodiode array detector (PDA) was used for the determination of phenylethanolamine A in animal feeds. The samples were treated by shaking extraction using different extract solutions (formula feed; 0.1% formic acid-methanol; concentrate feed; 0.1% formic acid solution-methanol (10:90, V/V); per-mixed feed; 0.1% formic acid solution-methanol (20:80, V/V) according to the characteristics of matrix, then purified by PCX solid phase extraction columns. A Waters Acquity UPLC BEH Shield RP18 column was used with 0.1% formic acid solution-methanol as mobile phase for gradient elution. The flow rate was 0.4 mL/min and the detection wavelength was 278 nm. Under optimum conditions, there was a good linear correlation (the calibration coefficient was above 0.999) in the range of 0.1-100.0 mu g/L. The limit of quantitative (LOQ) was 1.0 mg/kg. The recovery was in the range of 68.7%-96.8%, and the relative standard deviation of determination (RSD) was 2.4%-7.0% for three types of animal feeds. The method is simple and fast with wide linear range. It has been applied to analyze real samples with satisfactory results.
