Construction of transfer vector plasmid for herpes virus of turkey

摘要： In the present study, a transfer vector plasmid for herpesvirus of turkey (HVT) was constructed by insertion of LacZ gene cassette into SalI restriction site of glycoprotein A (gA) encoding region of HVT strain Fc-126. The LacZ gene, driven by SV40 early promoter, was employed as a reporter gene. Subsequently, chicken embryo fibroblasts(CEF) were cotransfected with the transfer vector plasmid and total DNA of HVT. On day 4 or 5 after cotransfection, some blue plaques appeared in presence of X-gal in agarose-overlay. The preliminary result suggests that the gA gene is nonessential for HVT replication in cell culture;and can be used as a insertion region for foreign gene.
