Characterization and impact of olfactory cues from aphid honeydew microbes on Harmonia axyridis

摘要: Microbial volatile organic compounds (mVOCs), produced by microorganisms associated with honeydew, play a crucial role in herbivore-natural enemy interactions. Nevertheless, the function of mVOCs on predatory ladybirds remains largely unknown. We hypothesized that the ladybird Harmonia axyridis (Coleoptera: Coccinellidae) could recognize and detect aphid honeydew-associated mVOCs for prey location. Our results showed that H. axyridis exhibited positive responses to the honeydew of Sitobion miscanthi (Hemiptera: Aphididae) and the mVOCs emitted by two honeydew-associated bacterial strains, Erwinia tasmaniensis MH4 and Klebsiella quasipneumoniae subsp. similipneumoniae MH6. A total of 16 mVOCs were detected by gas chromatography-mass spectrometry, with 3 mVOCs, namely 6-methylhept-5-en-2-one, 4-methylpentan-2-ol and 3-methylbutan-2-ol to be specific or significantly higher in attractive bacterial strains. Electroantennography and olfactory behavioral assays showed that H. axyridis prefers 6-methylhept-5-en-2-one and 4-methylpentan-2-ol. 3D modeling and molecular docking predicted that the H. axyridis olfactory binding proteins (HaxyOBP) HaxyOBP1 and HaxyOBP15 strongly interact with both compounds. Fluorescence competition assays further demonstrated that HaxyOBP15 exhibits a strong affinity for these attractive compounds. RNAi-mediated knockdown of HaxyOBP15 reduced H. axyridis' behavioral responses to 6-methylhept-5-en-2-one and 4-methylpentan-2-ol, confirming the critical role of HaxyOBP15 in detecting these mVOCs. We provide new insights on behavior of ladybirds in response to honeydew-associated mVOCs, contributing to research toward eco-friendly pest management methods targeting aphids.
