Development of Rc-targeted molecular marker for breeding anthocyanin-rich red rice

摘要: Red rice is characterized by high levels of anthocyanins, which have been recognized for their health benefits. In our previous research, we developed several homozygous lines with red pericarp, providing foundational materials for the enhancement of red rice through molecular-assisted breeding (MAS). In the current study, we utilized a single-nucleotide polymorphism within the Rc gene to develop a co-dominant derived cleaved amplified polymorphic sequence (dCAPS) marker, designated as Rc_dCAPS1. Utilizing Rc_dCAPS1, we successfully introgressed the Rc allele from the most representative donor lines, ZNS-#22 and X134-#2, into the male sterile line T710S and restorer line Xiang32. Rice seeds from five Rc-containing lines and their combinations were measured for anthocyanidins, and high accumulation levels of delphinidin and cyanidin were observed in these lines. Moreover, the five improved representative lines T71S-#1, T71S-#2, Xiang32-#1, Xiang32-#2, Xiang32-#3, and their combinations displayed some superior agronomic traits, such as increased plant height and panicle length, compared to their parents, respectively, suggesting that back-crossing selection positively influenced key agronomic traits. Furthermore, we found that these 5 improved lines showed markedly enhanced susceptibility to blast fungi compared with their respective wild parents. These results indicate that the optimized marker Rc_dCAPS1 is effective in introducing Rc through MAS. It is conceivable that our method could be applied to most white pericarp rice varieties and would greatly accelerate the breeding of new red rice varieties with restored recessive frame-shift alleles for crop improvement.
