Cyclic-di-AMP hydrolase GdpP is critical for stress response of Lactiplantibacillus plantarum WCFS1

摘要: To adapt to changes in the external environment, bacteria need to rely on second messenger signaling systems to respond quickly. As one of the second messengers, cyclic di-adenosine monophosphate (c-di-AMP) plays several key regulatory functions in this effort. To improve the stress resistance of probiotics in food processing and application, we explored the role of c-di-AMP hydrolase GdpP in the stress physiology of Lactiplantibacillus plantarum. Our results indicated that GdpP regulates many biological processes in L. plantarum. Its absence led to multiple bacterial defects such as growth inhibition, reduced biofilm formation, abnormal cell morphology, thinner cell wall, as well as compromised cell membrane properties. Systemic changes in these resistance determinants ultimately affected the stress response of L. plantarum, rendering it less tolerant to multiple stressors such as bile salts, osmolality, heat, oxidation, and antibiotics. In this study, we have confirmed the important contribution of GdpP and c-di-AMP to the stress resistance of L. plantarum and provided a new strategy for engineering probiotics to alter its properties.
