Assessing oxytetracycline stress and antibiotic resistance gene dynamics during anaerobic treatment of cow manure

摘要: Antibiotics and antibiotic resistance genes (ARGs) have become global issues. While most studies focus on the effects of antibiotics on anaerobic digestion (AD), systematic investigations into their subsequent management and application in biogas slurry remain limited. This study assessed the effect of Oxytetracycline (OTC) stress on methane production and ARGs response in continuous AD and subsequently optimized storage methods for biogas slurry maturation and soil applications. The results showed that across phases I to IV, the average daily methane yields were 204.44, 188.19, 133.98, and 92.19 mL/g*VS, respectively. With increased OTC stress, methane yield declines primarily due to the inhibition of dominant methanogen, especially Methanosarcina. Meanwhile, OTC stress induces the proliferation of ARGs. After 60 days of storage, the seed germination index (GI) of the biogas slurry exceeded 90%, indicating its plant safety. Compared to closed storage, the OTC content was reduced 1.5 times under open storage, as well as improved GI, making it more suitable for field applications. With open and closed storage of digestate, soil pH increased from 6.37 to 7.48 and 7.65, respectively, while organic matter rose from 9.18 to 9.36 and 10.13 g/kg. However, the abundance of ARGs in surface soil slightly increased, particularly with the application of slurry stored under closed conditions. These findings may help mitigate OTC and ARG pollution in anaerobic manure treatment.
