Genome-wide analysis of potato xyloglucan endotransglucosylase/ hydrolase (XTH) gene family revealed StXTH9 involved in drought stress responses

摘要: Drought is a common abiotic stress during plant growth and development, and XTH genes have been reported to be involved in a variety of abiotic stresses, but few studies have been carried out on drought-resistant XTH genes and molecular mechanisms in potato. In this study, 25 potato XTH genes were identified based on potato genome-wide data, with conserved structural domains (Gh16 and XET_C), universal functional structural domains and five conserved motifs with known functions. Evolutionary tree results showed that they could be classified into three major subfamilies. These StXTHs genes were distributed on 12 chromosomes of potato, with the highest number on chromosome 7; gene duplication analysis indicated that segmental duplication and tandem duplication might be the main reasons for the amplification of the XTH gene family in potato. Each StXTH gene has multiple promoter cis-acting elements involved in various regulatory mechanisms. Bioinformatics analysis screened 15 drought-related genes, and qRT-PCR analysis revealed that 15 XTH genes were involved in drought stress response; furthermore, we localised the StXTH9 gene in the cell wall, and heterologous transformation of tobacco revealed that the StXTH9 gene was involved in a negative regulatory role in drought stress response.
