Identification of a Vibrio cholerae Isolate as the Causal Agent of Ascites Disease in Cultured Mandarin Fish Siniperca chuatsi (Basilewsky)

摘要: The mandarin fish Siniperca chuatsi (Basilewsky) is an important commercial fish species in China and other countries because of its relatively high nutritional value. However, bacterial infection has become a major economic problem in its farming. In this study, a virulent strain was isolated from diseased mandarin fish suffering from ascites disease and identified as a Vibrio cholerae isolate using the ARIS 2X system. Named GYL, its taxonomic position was determined by a nucleotide BLAST search on the NCBI website and phylogenetic analysis. A phylogenetic tree, constructed using the neighbor-joining method, showed that the GYL isolate was the V. cholerae strain (GenBank accession no. JQ686623), previously isolated from diseased freshwater fish. Three genes encoding cholera toxin transcriptional activator (toxR), outer membrane protein (ompU), and hemolysin (hlyA) were present in the GYL isolate, further confirming its potential virulence. The GYL isolate was susceptible to enrofloxacin, gentamicin, norfloxacin, ofloxacin, streptomycin, and tetracycline, and resistant to trimethoprim. This infection represents an emerging threat to mandarin fish farming that should attract more attention due to the potential pathogenicity of V. cholerae in mandarin fish diseases.
