Transcriptomics and EtSRG1 Functional Analyses Provide New Insights Into the Regulatory Mechanisms of Secondary Appressoria Formation of Exserohilum turcicum

摘要: Northern corn leaf blight (NCLB) significantly reduces maize yields globally and leads to substantial economic losses. Exserohilum turcicum, the causal agent, produces multiple secondary appressoria during the infection of maize leaves after a failure of the primary appressoria. However, the regulatory mechanisms of the formation of multiple appressoria remain unclear. In the present study, using the artificial substrate Parafilm, we observed that the multiple secondary appressoria were continuously produced until 96 h post-inoculation (hpi), with the newly emerging secondary appressoria demonstrating penetrative ability. Transcriptomic analyses of appressoria showed that new energy reserves and the glycolytic pathway were both involved during secondary appressorium formation. Additionally, proteins associated with the stress response and autophagy were found to play a role in this process. Comparative analysis of gene expression profiles at 2, 4 and 8 hpi identified 47 upregulated differentially expressed genes (DEGs), including EtSRG1, containing an NmrA-like domain. Sporulation levels in three EtSRG1-silenced transformants were significantly lower than those in wild-type strains, and conidia from these mutants exhibited rare germination. These findings indicate that EtSRG1 is crucial for the sporulation of E. turcicum. Overall, this study provides new insights into the energy utilisation and molecular regulatory mechanisms involved in the formation of secondary appressoria in E. turcicum, potentially contributing to the development of novel control strategies.
