Pharmacokinetics, Tissue Distribution, and Elimination of Flumequine in Japanese Seabass (Lateolabrax japonicus) following Intraperitoneal Injection and Oral Gavage

摘要: The pharmacokinetics, tissue distribution, and elimination of flumequine were investigated following intraperitoneal injection and oral gavage at doses of 20 mg/kg in Japanese seabass (Lateolabrax japonicus) in 25 degrees C sea water with 26 parts per thousand salinity. Following a single intraperitoneal injection, the profile of plasma concentrations vs time was best described by a two-compartment open model with first-order absorption. The distribution and elimination half-lives (t1/2a and t1/2 beta) were 0.71 h and 8.163 h, respectively. Total body clearance (CLs) and apparent volume of distribution (Vd) were 256.0 ml/h/kg and 3.012 l/kg, respectively. The plasma concentration-time curve after oral gavage (force feeding) did not fit the compartment model and plasma parameters were calculated by a non-compartment model based on statistic moment theory. The area under the concentration-time curve from 0 to 8 h (AUC0-infinity), mean residue time from 0 to infinity h (MRT0-infinity), half-life (t1/2z), and body clearance (CLz) were 26.40 mu g(h/ml), 21.97 h, 14.90 h, and 757.0 ml/h/kg, respectively. The plasma protein binding rates in vivo were 13.4% at 1 h and 31.3% at 12 h following intraperitoneal injection, and 19.3% at 4 h and 45.2% at 18 h after oral gavage. Muscle and liver parameters were calculated by statistic moment theory following intraperitoneal injection and oral gavage. After intraperitoneal injection, AUC0-infinity, MRT0-infinity, and CLz were 73.99 mu g(h/g), 9.86 h, and 270.0 g/h/kg for muscle and 140.54 mu g(h/g), 22.46 h, and 142.0 g/h/kg for liver. After oral gavage, AUC0-infinity, MRT0-infinity, and CLz were 9.17 mu g(h/g), 37.62 h, and 2181.0 g/h/kg for muscle and 60.91 mu g(h/g), 45.80 h, and 328.0 g/h/kg for liver. Results indicate that flumequine is rapidly distributed to tissues and quickly eliminated from muscles when administrated to Japanese seabass.
