Molecular identification and characterization of pig's Cdh16 gene

摘要: Cdh16 (Cadherin16, or Ksp-cadherin) is strongly expressed in epithelia of nephron tubules and collecting ducts in rabbit, mouse and human. Transgenic mice with exogeneous gene driven by Cdh16 promoter (e.g. Ksp-Cre) is widely used in basic research of nephrology. To determine whether the pig Cdh16 gene could be utilized to generate transgenic pigs with kidney-specific expression pattern just like the mouse Cdh16 gene, we cloned pig Cdh16 gene, analyzed its expression profile and promoter activity. Pig Cdh16 gene has 18 exons, the same number as human, mouse and rabbit's. Meanwhile, pig cadherin16 protein shows over 77% identity with its homolog from human, mouse and rabbit, and they all have similar protein domains. Kidney is the organ in which pig Cdh16 is transcribed at the highest level, but there is also low transcription in pig's lung, too. In pig kidney, IHC staining reveals that cadherin16 is localized on the basolateral membrane of epithelial cells of nephron tubules and collecting ducts, which is mostly in consistent with human, mouse and rabbit's cadherin16 protein. Furthermore, the 5' flanking region of pig Cdh16 gene has promoter activity in LLC-PK1 cell line, an epithelial cell line derived from porcine proximal tubules. In summary, Cdh16 gene and its protein product are highly conserved among human, mouse, rabbit and pig. The promoter of pig Cdh16 gene could be used to drive transgene expression in pig kidney.
