The inhibitory mechanism of perillaldehyde against Penicillium digitatum and its role in delaying citrus green mold

摘要: Green mold, as a major postharvest disease in citrus caused by Penicillium digitatum (P. digitatum), can lead to severe rot and considerable economic loss. This study aimed to explore the antifungal properties of perillaldehyde (PAE) against P. digitatum, and to further elucidate its underlying mechanisms. PAE showed excellent inhibition ability against P. digitatum, whose minimum inhibitory concentration and minimum fungicidal concentration against P. digitatum were 0.625 mL L-1 and 1.25 mL L-1, respectively. Meanwhile, PAE suppressed the mycelial growth via folding, twisting, and shrinking mycelium of P. digitatum, as well as inhibited its spore germination and germ tube. Otherwise, PAE altered the cell membrane permeability and integrity, which in turn led to cellular contents leakage, the reactive oxygen species accumulation, and lipid peroxidation. Metabolomic analysis showed that PAE mainly affected alpha-linolenic acid metabolism, steroids degradation and steroids hormone biosynthesis pathways, thus disturbing the growth of P. digitatum. Finally, in vivo experiments successfully confirmed that PAE could decrease the lesion diameter and decay rate in citrus during storage. Hence, this study provides new insights into understanding PAE as a natural antifungal agent for regulating green mold in postharvest citrus.
