Molecular identification and recombinant expression of red sea bream (Chrysophrys major) hepcidin gene

摘要: Hepcidin is a member of antimicrobial peptides (AMPs) family. However, little is known about the structure and function of hepcidin in lower vertebrates such as fish. We have recently cloned a cDNA encoding hepcidin from cDNA library of spleen of red sea bream (Chrysophrys major) by EST analysis (Chen et al., 2005). In order to study the function of hepcidin in fish, an expression cassette harboring red sea bream hepcidin cDNA was constructed by using secretory plasmid pPICZaA and recombinant hepcidin was generated in yeast (Pichia pastoris). SDS-PAGE of the yeast cultures demonstrated that recombinant hepcidins were produced in the recombinant yeast. The supernatant from recombinant yeast culture exhibited a dose-dependant antibacterial activity on the growth of E. coli DH5a, E. coli ATCC25922, Pseudomonas aeruginosa and Aeromonas hydrophia.
