Immunomodulation of thymus by Cistanche deserticola polysaccharides analyzed via transcription-associated proteomic techniques

摘要: The aim of this study was to investigate the mechanism of immunomodulatory effects of Cistanche deserticola polysaccharide (CDP-D2-N1) on cyclophosphamide (CTX)-induced immune injury in mice. A model of CTXinduced immune injury in mice was established, and the effects of CDP-D2-N1 on body weight, thymic index, cytokines, oxidative stress products and oxidoreductase were evaluated. Meanwhile, transcriptomics and proteomics were applied for joint analysis. The results showed that CDP-D2-N1 inhibited body weight loss, increased organ index, enhanced the expression of anti-inflammatory factors, and increased the activities of GR, SOD, and CAT enzymes in immune injury mice. Through the multi-omics study, CDP-D2-N1 reversed CTX-induced immune injury, promoted functional recovery of Th17 cells, and produced immunomodulatory effects on mouse thymus. This finding provides a theoretical reference for the development of polysaccharides as natural immunomodulators.
