IMPROVEMENT ON THE THERMOSTABILITY AND ACTIVITY OF APX1 FROM ENERGY PLANT JATROPHA CURCAS L. BY HYPER-ACIDIC FUSION PARTNERS

摘要: Cytosolic APX1 (ascorbate peroxidase 1) prevalently plays a crucial role in the response and tolerance to numerous environmental stresses and acts as the key regulator of the cellular redox state in plants. Nevertheless, APX1 is thermolabile and prone to be inactivated even under moderate heat stress, which was proposed as a causative factor for plant heat-susceptibility. In this study, we cloned the APX1 gene (JcAPX1) from the energy plant Jatropha curcas L. The recombinant JcAPX1 and its C-terminal fusions with hyper-acidic peptides were evaluated for their thermostability and activity, through SDS-PAGE and in gel activity staining plus spectrometric enzymatic assays after a heat treatment of gradient temperature (37 degrees C similar to 57 degrees C)for 30min. Our results demonstrated that the hyper-acidic fusion partners could efficiently improve the thermostability and activity of JcAPX1, with an elevated heat tolerance of at least 2 degrees C higher than the original. Presumably, this study is of a considerable potential to enhance plant heat tolerance, not only applicable for energy plant Jatropha curcas L., but also for other plant species especially of economic significance but inherent thermolability.
