Qualitative Analysis of Unknown Color Additive by Liquid Chromatography-Ion Trap-Time of Flight Tandem Mass Spectrometry

摘要: Some unknown color additive used for coloring the skins of oranges was qualitative analyzed by UV. Vis spectrophotometry coupled with ultra-fast liquid chromatography-diode array detector-ion trap-time of flight tandem mass spectrometry (UFLC-PDA-MS-IT-TOF) and identificated by ultra performance liquid chromatography. tandem mass spectrometry (UPLC-MS/MS). The characteristic wavelength of the unknown color additive was obtained by UV-Vis spectrophotometry, and it was used in the experiment to show the retention time by PDA detection and to differentiate from other disruptors by mass spectrometry. When the unknown color additive was separated from complex matrix, the m/z and the fragment ions from MS1 to MS5 were obtained, and based on the MS/MS fragments and accurate masses of the target compounds, the MS fragment path was be induced. According to all the data and the internet information, the unknown color additive that was used for coloring the skins of oranges was "Citrus red No. 2" whose formula was C18H16N2O3 and m/z was 309.1932. UPLC-MS/MS was used to test the qualitative result. By comparing the unknown color additive with citrus red No. 2 standard, its retention time, ion. pairs m/z 309.3>153.3 and 309.3>278.3 and ion ratios were the same. The results were confirmed accurate. This work provides an effective ideas and methods for the unknown compounds qualitative.
