Influence of chemical crosslinker SPDP on the spectrum and introduced pyridin-2-thione moieties of R-phycoerythrin

摘要: R-phycoerythrin (RPE) purified from Polysiphonia urceolata with absorption maxima are at 498, 540, 565 nm and a fluorescence emission maximum at 575 nm. RPE is an excellent fluorescent dye because of its high fluorescence quantum efficiency and large Stokes shift. RPE must first derivated by chemical crosslinker such as SPDP then conjugated with antibodies or other proteins into a fluorescencetracer. The amount of the introduced derivative groups and the structure and function of RPE were both be considered during chemical derivation. The influence of different concentrations of SPDP on the absorption and fluorescence spectra and amount of the introduced pyridin-2-thione moiety of RPE was the first investigated. The results showed that the absorbance and fluorescence intensity both slightly decreased with the increase of the molar ratios of SPDP to RPE, however, Mole number of molecules of pyridin-2-thione introduced into RPE increased as the molar ratio of SPDP to RPE increasing within the first 1.5 hours from derivation, then sustained stable values after 2 hours. The appropriate molar ratios of SPDP to RPE were from 20 to 300 based on the spectrum and pyridin-2-thione derivatives, and the appropriate derivation time was 2 hours.
