Construction of cDNA Library and Analysis of SSRs in ESTs of Litchi (Litchi chinensis Sonn.)

摘要: A high-quality cDNA library was built with two subsections representing the cell division and cell enlargement periods of litchi pericarp fruit development. These subsections were sequenced, revealing 1353 cell division unigenes and 1331 cell enlargement unigenes. ORF prediction, functional annotation, functional classification, and bioinformatics analysis of these unigene sequences were performed, revealing 308 and 387 homologous genes in each subsection. The 1331 unigenes from the cell enlargement cDNA library were used to search for SSR sequences. A total of 220 SSRs distributed across 189 unigenes were found, accounting for 16.53% of the unigenes generated in this study. They consisted of repeat units of mono-(>= 20), di-(>= 6), tri-(>= 5), tetra-(>= 4), penta-(>= 3), and hexa-nucleotide(>= 3). The average length and distribution distance of the EST-SSRs were about 18.43 and 4.42 kb, respectively. Di- and tri-nucleotides were the most common types of repeats, and they occurred at similar frequencies, accounting for 69.09% of all the SSRs. 72 motif types were observed in all the SSRs. The most frequently observed motif was GA/TC (16.82%), followed by AG/CT at 4.55%, A/T at 11.82%, AT/TA at 5.00%, and GAA/TTC at 3.64%.
