Sequence analysis and preliminary study on the correlation with ovulation of the cyp19a1a gene in Hemibagrus guttatis

摘要: The full-length CDS sequence of cyp19a1a cloned from the ovary of Hemibagrus guttatis was 1587 bp, encoding a 528 aa protein with a conserved cytochrome P450 family 19 domain. Multiple sequence alignment and phylogenetic analysis revealed that Cyp19a1a of H. guttatis was most closely related to that of Hemibagrus wyckioides and clustered with other teleost fish. H. guttatis cyp19a1a was found to be specifically and highly express in the ovary, mainly in granulosa cells and theca cells. Our study demonstrated that elevated density and reduced water exchange rates significantly downregulated ovarian cyp19a1a expression and impaired ovulation rates in female H. guttatus. Conversely, E2 treatment exhibited a dose-dependent biphasic effect, enhancing cyp19a1a levels at 10 mu g/kg but suppressing both expression and ovulation at 80 mu g/kg. Higher aquaculture densities were associated with decreased cyp19a1a expression and ovulation rates, while increased water exchange rates correlated with elevated cyp19a1a expression and higher ovulation rates. E2 treatment exhibited a dose-dependent suppression of ovulation in H. guttatis, while 5 mu g/kg and 10 mu g/kg reduced ovulation rates by 30 %, higher doses completely inhibited ovulation. These findings provide insights into the regulation of cyp19a1a and its impact on the ovulation performance of H. guttatis.
