Isolation, Sequence Identification and Tissue Expression Profile of a Novel Pepper Gene-DFR

摘要: The complete CDS sequence of "Yunnan Purple Pepper No.1" gene dihydroflavonol 4-reductase (DFR) was amplified using the reverse transcriptase polymerase chain reaction based on the conserved sequence information of the tomato or other Solanaceae plants and known highly homologous pepper ESTs. The nucleotide sequence analysis of these two genes revealed that pepper DFR gene encodes a protein of 382 amino acids that has high homology with the PLN02650 protein of six species Nicotiana tabacum (100%), Solanum tuberosum (100%), Petunia x hybrida (92%), Gynura bicolor (90%), Centaurea maculosa (87%), Rhododendron simsii (86%). The tissue expression analysis indicated that the pepper DFR gene was moderately-expressed in pericarp and leaf, weakly in stem, flower, placenta and seed, and hardly expressed in root. Our experiment established the primary foundation for further research on the novel pepper gene.
