Relationships between abamectin resistance and the activities of detoxification enzymes in the cotton bollworm, Helicoverpa armigera

摘要: To understand the mechanism of abamectin resistance in Helicoverpa armigera (Hubner), abamectin resistance was selected for and detoxification enzymes were quantified for 27 generations of the insect. With continuous selection pressure, resistance of H. armigera to abamectin increased slowly from generation F(0) to F(21) and then increased rapidly from generation F(21) to F(27). Resistance of F(21) and F(27) larvae was 121 and 822 times greater, respectively, than that of F(0) larvae. The plot of abamectin resistance on numbers of generations subjected to abamectin-selection pressure produced a sigmoid curve. Selection for abermectin resistance had no effect on acetylcholinesterase (AChE) activity but increased the activities of carboxylesterase (CarE) and glutathione S-transferases (GST) in H. armigera larvae. Relative to activities in F(0) larvae, the activities of CarE and GST in H. armigera larvae after 27 generations of abamectin-selection pressure increased 2.5-fold for alpha-NA CarE, 2.8-fold for beta-NA CarE, and 2.1-fold for GST. Treatment of F(27) larvae with piperonyl butoxide (Pb), which inhibits the activity of microsomal mixed-function oxidases (MFOs), increased the toxicity of abermectin by 6.7-fold. These results suggest that the resistance of H. armigera to abamectin could be associated with MFOs, GST and CarE, but not with AChE.
