IMPROVED MYCELIUM BIOMASS BY SUBMERGED CULTURE OF ANTRODIA CINNAMOMEA USING PLACKETT-BURMAN DESIGN AND RESPONSE SURFACE METHODOLOGY

摘要: Antrodia cinnamomea (A. cinnamomea) fruiting body cultivation has hardly been developed successfully in solid-state fermentation. Thus, to improve its biomass, response surface methodology with a Box-Behnken design was used to predict the optimal conditions in a submerged culture. The most influencing factors of A. cinnamomea biomass (Dry weight, DW) were glucose, malt dust, yeast extract, V-B1 and chitosan (p < 0.05). The predicted value of biomass after culturing for 15 days was 1.7843 g/100 mL (DW) under 35.16 g/L glucose, 42.86 g/L malt dust, 20.80 g/L yeast extract, 0.39 g/L VB1 and 0.85 g/L chitosan conditions, while the actual biomass yield was 1.6765 +/- 0.0103 g/100 mL, which was 2.5 fold more than the control. There was also a significant relation (p < 0.05) between the biomass and triterpenoid concentration. Chitosan was thought to have enhanced the growth rate of mycelium and triterpenoid production, and as a result could be used as a natural additive in submerged culture.
