DNA fingerprints of macadamia constructed using genome-wide distributed SNP markers

摘要: Macadamia is an important economic fruit tree widely cultivated in tropical and subtropical regions, but the lack of DNA fingerprints hinders the efforts to improve the identification efficiency of macadamia germplasm resources. This study aims to construct DNA fingerprints for the identification of macadamia germplasm resources based on genome-wide SNP markers. Genome-wide SNP markers were developed based on whole genome re-sequencing data of 187 macadamia germplasm resources from the United States of America, Australia, South Africa, Israel and China. After rigorous filtering, 284 SNP markers obtained from nearly 124 million SNP markers formed the core marker set with moderate polymorphism. Based on the 284 core SNP markers, DNA fingerprints of the 187 macadamia germplasm resources were constructed and two-dimensional barcodes containing the genetic information of each germplasm resource were generated. The SNP fingerprints constructed in this study provide accurate and reliable fingerprint codes for the identification of macadamia germplasm resources, and contribute to the subsequent applications such as authentication and intellectual property protection of macadamia germplasm resources.
