In vitro induction and characterization of octaploid Schizachyrium scoparium, an ornamental grass

摘要: Schizachyrium scoparium is a kind of ornamental grass that has been planted in China and abroad. In this study, the calli of tetraploid S. scoparium was used as the material and colchicine was used as the inducer of polyploid. The effect of different treatment concentration and time on polyploid induction was studied. The induction system of S. scoparium octaploid was established. The results provide a novel avenue for germplasm innovation and breeding of new cultivars of S. scoparium. The difference between the two plants was compared by stomatal parameter analysis, flow cytometry analysis, chromosome counting, growth parameter and phenological analysis. The optimal colchicine treatment involved exposure to 0.2% colchicine for 72 h. The highest percentage induction of octaploid plants was 26.7%, resulting in the regeneration of 48 independent octaploid individuals from calli. Notably, octaploid plants exhibited morphological changes compared with tetraploids, including a reduction in stomatal density and an increase in stomatal size. Plant height was reduced by 50% and leaf width was significantly increased. The average number of spikes per plant was reduced by 8.7 and flowering was delayed by 21 d. These findings suggest that the induced octaploid plants have potential as a valuable resource for germplasm innovation for new cultivars of S. scoparium.
